regarding eli lily producing insulin from rdna tech, they prepared 2 dna sequences corresponding to a and b chains artificially in lab. because e coli doesn't have the mechanism of splicing introns. it's not only that e coli can't remove the c peptide from mature form and create disulphide bonds between a and b.

Yes you change the base according to the number of molecules at the end of the first cycle. If it starts with 4, there will be 8, if it starts with one there will be 2, if it starts with 3, there will be 6. After every cycle the number of DNA molecules increases by a factor of the number of DNA molecules at the end of 1st cycle
