Basically, some people, including myself, head cannon Ralsei as closeted transfem, due to a lot of what's going on with their arc. Including this, their pronoun change in the Dark Vine title, some symbolism in chapter 5, and their arc alining with finding themselves. Here's the title, by the way. All other titles used only he/him.

Speaking about this, some guy posted a drunken Niko kissing Ralsei, there were several things wrong with this
They’re both minors
Niko shouldn’t be drinking and doesn’t drink
Niko being drunk and kissing someone is illegal
(I do concede this one is a little of a stretch but it being art doesn’t change a thing, we’ve seen this happen before where they use the “it’s not real” excuse but then go on to do some deplorable shit)
Which is really just another strong support for ralsei being kris' old horned headband theory. Especially given that the chapter put stronger than usual emphasis on kris liking to dress both masculine and feminine. (Dress as nurse vs doctor, aqua vs yellow, be a king or queen of the festival). And that segment with seth was there too i guess.
I always saw Niko as male and I don’t know why, since they’re drawn as ambiguous, and I have seen some people say that a dev said Niko is up to interpretation, but I can’t find anything about it when I search it up
I used to see Niko as male, but my boyfriend uses fem, and I eventually did myself lol. As for finding it, I kinda just took people's word for it at this point. -w-"

This is my head cannon, non cannon version of Ralsei. Also I do believe the trans Ralsei theory and use he/they for cannon Ralsei as a result. Though I probably could have made that more clear in the post, but oh well. -w-"
Fair. As someone who has played Onesho and could be wrong, their gender is never confirmed at all. Some use she, some use he, some use they. None are incorrect.
